Según sus propias estimaciones, Harry Haley McClaskey grabó cerca de 12.000 discos, logrando una enorme popularidad en el periodo comprendido entre 1903 y 1928, con un total de 15 números uno en su carrera en solitario. Fueron igualmente celebrados los duetos junto a cantantes como Albert Campbell, Frank Stanley, y otros, así como sus colaboraciones en conjuntos vocales, como el Columbia Male Quartet, el Peerless Quartet y el Sterling Trio.
Harry comenzó a cantar desde los cinco años en su St. Stephen natal, y a los trece cantaba como tenor en la Artillery Band de Saint John, ciudad a la que se había trasladado su familia. Su primera actuación la Opera House de St. John llegó el 14 de abril de 1901, junto a la soprano escocesa Jessie MacLachlan. Fue descubierto, el 30 de septiembre de 1901, por Giuseppe Campanari, barítono de la Metropolitan Opera, que lo animó para que tomara clases de canto en Nueva York, hacia donde viajó en 1902.
Cantó con el coro de la Grace Methodist Episcopal Church, y entre sus maestros estaban John Dennis Meehan (o Mehan) y Kate Stella Burr, de quien adoptaría su nombre artístico, que usó ya desde sus primeras grabaciones para Columbia Records, este mismo año. Burr llegó a Columbia en un momento crucial, ya que su tenor estrella, George J. Gaskin, estaba en los últimos años de su carrera.
Pero Burr grabó igualmente en otras compañías. Para Edison Records comenzó a grabar en noviembre de 1904, bajo el nombre de Irving Gillette, compañía en la que se mantuvo en activo hasta octubre de 1914, y con Victor Records sus primeras sesiones de grabación tuvieron lugar el 4 de enero de 1905.
Burr demostró ser un artista de enorme éxito, especialmente en la década de 1910, convirtiéndose en el cantante que más números uno acumuló durante estos años. Su grabación más famosa fue "Just a Baby's Prayer at Twilight", que vendió un millón de copias. Se estima que de sus grabaciones se vendieron alrededor de 240 millones de copias.
En 1906 Burr se unió al Columbia Male Quartet, bajo la dirección de Frank C. Stanley, como segundo tenor. Más tarde, cuando se mudaron al sello Victor, fueron conocidos como el Peerless Quartet, y cuando Stanley murió, en 1910, Burr se hizo cargo de la dirección del grupo, que continuó (con varios cambios de personal y de nombre a lo largo de los años) como popular grupo de grabación, y en directo, hasta 1928, cuando se disolvió. Burr también fue miembro del Metropolitan Trio y del Manhattan Mixed Trio, ambos con Frank C. Stanley y Elise Stevenson.
A fines de la década de 1920 la carrera discográfica de Burr había terminado. Las tecnologías de grabación eléctrica estaban favoreciendo a los tenores de estilo crooner, como Gene Austin y Al Bowlly, pero gracias a las ventas de sus discos, Burr logró amasar una importante fortuna. Como hombre de negocios tuvo altibajos: su incursión en el mundo de las discográficas fue un rotundo fracaso, pues Paroquette Records, fundada junto a Fred Van Eps, se mantuvo en activo únicamente entre 1915 y 1917.
Mejor suerte tuvo en el mundo radiofónico, donde había debutado en 1920. En 1928 fundó la productora Henry Burr, Inc., produciendo numerosos programas para cadenas de radio comerciales en la década de 1930.
En octubre de 1929 perdió una parte sustancial de su riqueza tras el desplome de Wall Street, pero al mes siguiente fue nombrado director de la Oficina de Artistas de la CBS, que acababa de organizarse bajo la propiedad de William S. Paley.
Alrededor de 1935, volvió a actuar en la radio como miembro de la compañía WLS Chicago National Barn Dance, que se transmitía por NBC los sábados por la noche. Pronto se convirtió en un actor destacado en el programa, en el que permaneció durante cinco años hasta poco antes de su muerte.
